Chronic inflammation is a major causative factor involved in practically every illness. It is the driving force not only of chronic pain but also the pathogenesis of heart disease, cancer, arthritis, colitis, PMS osteoporosis, neuro-degenerative diseases (such as Alzheimer’s) and most other degenerative diseases.

There is a pro-inflammatory cell-signaling molecule called Nuclear Factor Kappa B (NF-kB) that resides in the cytoplasm of the cell. It remains inactive if it remains in the cytoplasm bound to its inhibitor.
However, when injurious and/or inflammatory stimuli occur (such as ATP depletion and oxidative free radical stress), NF-kB is released from its inhibitor and moves into the cell’s nucleus where it activates the genes responsible for increasing the activity of pro-inflammatory enzymes which increase the levels of pro-inflammatory eicosanoids (PGE2, LTB4) and cytokines (IL-1, 6 TNF).
Although the subject is complicated, having a simplified understanding is all you really need. This is depicted by the graphic below. Just know that two of the main drivers of chronic inflammation are ATP depletion and Free radicals (oxidative stress).
ATP synthesis and antioxidant defense work together in synergy. ATP energy is necessary for glutathione (GSH) synthesis which is the cell’s main antioxidant and without sufficient ATP, the other cellular antioxidant defense processes also break down.
Adequate ATP synthesis is dependent on adequate macronutrients (glucose, amino acids, etc.) and a wide variety of micronutrients (vitamins, minerals and trace minerals) for optimum performance. These nutrients must be bio-available (absorbed) and in their biologically active forms.
Without antioxidant protection, polyunsaturated fatty acids can oxidize. This causes activation of NF-kB that up-regulates the gene coding for the pro-inflammatory enzyme production which subsequently increases all the inflammatory mediators (IL1,6; TNF, PGE2 and LTB4).
